Put a vacuum pump on the fuel regulator and see if it 'holds' a vacuum. It should.

This is for your 1978. Fuel pump must not be working or is fuel line is plugged.
Test fuel pump cranking and with AFM (air flow meter) flap open

Check fuel pressure.
It runs when You "put some gas in the throttle body and it start little bit hard to start but run for 4-5 sec"
It has to be a fuel problem.

Do you have White wire going from (N) Coil #1 to (J21) ECM #1 (electronic control module)?

Depends on which injection system you have in the bus.
Some injection systems came with a Mechanical fuel pressure regulator.
Some injection systems came with a Vacuum operated pressure regulator.

What is the number on your ECU (electronic control unit)
thnx to mayor ratwell

Keep it simple. When a fuel injected bus suffers starting problems, you would do well to remove the spark plugs, clean each electrode firing surface, re-gap, and then get the engine running and keep it running. Slightly sooty plugs will get "wet" and ground themselves out through the carbon.

The fuel pressure regulator has a check valve that holds back the fuel in the rail if it is under 1 bar.

Sooooooooooo....?
Are you saying "wow, it now drives as if it's brand new", or "wow, I have a brand new gauge"?
